Platelet surface expression (% of positive cells) of P-selectin and fibrinogen binding were measured in unstimulated samples (basal activation) and after stimulation with ADP 0.1-10 μM for 20 minutes before fixing with 0.2% paraformaldehyde. For platelet sensitivity to PGI₂, samples were pre incubated with PGI₂ 1, 10, and 100 nM for 90 seconds before; stimulation with ADP 10 μM and fixing at 20 minutes for P-selectin expression; or stimulation with ADP 1 μM and fixing at 5 minutes for fibrinogen binding.
